Sirolimus-eluting stent coated with biodegradable polymer by ultrasonic spray method and its controlled release |

One of the solutions about in-stent restenosis and late thrombosis is to control eluting of drugs, e.g. sirolimus (SRL), that can prevent tissue hyperplasia followed by vascular injury during implantation of a coronary stent. The SRL-loaded polymer solution was then sprayed onto the stent surface using ultrasonic spray. Polymer solubility parameter was introduced in this study to control roughness and thickness of polymer coated on SS substrates as varying conditions of solvent system. Release profiles, thermal properties, and compositions were measured by HPLC, DSC, and FTIR, respectively. FE-SEM images demonstrated that the thickness of the polymer layer was controlled up to 5 m. After SRL was loaded with 100 μg/cm2, the release behavior was controlled by top-coating and solvent system. Depending on the thickness and/or type of polymers, top-coating on drug-in-polymer layer produced significantly different profiles of SRL release. Solvent system influenced the release kinetics. |
