Polyethylenimine(PEI) has been known as an efficient non-viral vector due to highest cationic charge potentials. However, it has drawbacks such as its cytotoxicity. Many researchers tried to develop the less toxic PEI after chemical modification. In this study, low molecular weight PEI (10 K) was modified with urocanic acid (UA) which has the imidazole ring (PEIU). The PEIU was prepared by amidation of the amine of PEI with urocanic acid by using an 1-ethyl-3-(3-dimethylaminopropyl) (EDAC)/ N- hydroxysuccinimide(NHS) condensation system. The PEIU/DNA condensation was confirmed by electrophoresis on 1.0 % agarose gel. Particle sizes were determined using a light scattering spectrophotometer (ELS 8000, Otsuka Electronics, Ltd., Osaka, Japan). Cytotoxicity test was used by Cell Proliferation Assay Kit ( Promega Co.). The in vitro transfection experiment was checked by luciferase assay with several cells. The PEIU will be expected to be useful for clinical application of gene therapy with improving the transfection efficiency and cell viability.
