제목 | MePEG-PCL diblock copolymer containing DOPE-DNA complexes for gene delivery |
초록 | Recently numerous techniques have been introduced to deliver a foreign DNA into cells to alter their function. These techniques involved two major groups, viral and non-viral vectors. In general, viral vectors are having very efficient transfection ability. However some limitations can be occurred in their use. Viral vectors may provoke mutagenesis and carcinogenesis. Repeated administration of a viral vector induces an immune response which abolishes the transgenic expression. So the potentials and benefits of non-viral vectors such as cationic lipid or cationic polymer have been researched to overcome problems associated with viral gene delivery. In this study we prepared MePEG/PCL diblock copolymers to examine the possibility of these polymer micelles as gene vehicles. Negatively charged plasmid DNA and positively charged DOPE formed DNA complexes through electrostatic interactions. These hydrophobic complexes can be loaded into the polymer micelles. Due to PEG part, this micelle is more stable in blood stream than native DNA and can release DNA complexes in sustained manner. Transfection ability was investigated by luciferase assay on NIH-3T3 cells. And in order to evaluate cytotoxity of MePEG/PCL nanosphears, MTT test was preformed. |
