제목 Functional Cartilage Formation using Surface-Modified PLLA Scaffolds in Hydrodynamic Bioreactor System
초록 An optimal three-dimensional (3D) culture condition is essential to grow a hyaline-like cartilage tissue in vitro. It would allow chondrocyte to maintain their phenotype and produce cartilage-specific extracellular matrix (ECM). As compared to static culture, major benefits are found with bioreactors that provide cells with efficient mass transfer and controlled shear stress. In this study, rabbit chondrocytes were seeded onto various surface-modified PLLA scaffolds and the constructs were cultured in bioreactor system. The scaffold was grafted with acrylic acid (AA) using in situ direct plasma treatment method and then followed by RGD peptide grafting. Cultured for specific time periods, specimens were analyzed for cell numbers, viability, ECM contents, histology, and gene expression of ECM molecules. Different levels of hydrostatic pressure and shear stress were applied and their effects were evaluated in terms of the quality of the cartilage tissues formed in the bioreactor system.
