In this study, we synthesized poly(amidoamine) dendrimers conjugated with 6-aminohexanoic acid (Ahx) and l-arginine (Arg) by using HOBt/HBTU coupling method. 4th generation of PAMAM dendrimers was used because they display relatively high transfection efficiency and low cytotoxicity than other generations. In our previous study, it was confirmed that l-arginine grafted PAMAM dendrimers showed enhanced tranfection efficiency. 6-aminohexanoic acid was selected as a spacer molecule to increase hydrophobicity and to introduce the flexibility of the conjugated amino acids. The synthesized PAMAM derivative was analyzed by H1-NMR and agarose gel retardation assay was performed to confirm complex formation of polyplex. To analyze the mean diameter of polyplexes, dynamic light scattering method was used. And the in vitro evaluation of transfection efficiency and cytotoxicity of synthesized PAMAM derivatives, luciferase expression assay and WST-1 assay was performed, respectively. |
