제목 HYDROPHOBIC MODIFICATION OF L-ARGININE GRAFTED PAMAM (POLYAMIDOAMINE) DENDRIMER AS A GENE DELIVERY CARRIER.
초록 In this study, 6-aminohexanoic acid and γ-aminobutiric acid(GABA) were respectively conjugated to L-arginine grafted polyamidoamine dendrimer 4(PAMAM G4) as a spacer molecule. Comparatively high transfection efficiency of Arginine conjugated PAMAM was verified through our previous study. The 4th generation of PAMAM dendrimer was selected because of its relatively good transfection efficiency and low cytotoxicity compared to other generations. As a spacer molecule, 6-aminohexanoic acid and γ-aminobutiric acid were used to increase hydrophobicity and to introduce the flexibility of the conjugated amino acids. It is also try to recognize effect of space molocule's lenth by comparing 6-aminohexanoic acid and γ-aminobutiric acid. These synthesized PAMAM conjugates were analyzed by 1H-NMR. PAMAM-Ahx-Arg and PAMAM-GABA-Arg were examined the transfection efficiency by luciferase assay and protein assay. These compounds were also investigated cytotoxicity by WST-1 assay.
