제목 Amphiphilic Comb-like Poly(oxyethylene)s with Sulfide Linkages : Synthesis, Characterization, and Application for Drug Delivery System
초록 Amphiphilic polymers have been used widely as drug delivery vehicles because they have several useful properties for such applications (1) they can dissolve hydrophobic drugs, (2) they can form small-sized particles, (3) their supramolecular structures have thermodynamically stability, (4) controlled drug release is possible, and (5) rethiculoendothelial uptake can be prevented. However to use these polymers in practical drug delivery application, following criteria should be satisfied. First, the hydrophilic-hydrophobic balance (HLB) should be tunable to match amphiphilic drug carriers with the specific hydrophobicity of a drug, thus optimizing the drug-carrier interaction. Second, the drug carriers must be biodegradable and easily extracted from the body. Third, the polymers themselves should not cause any harmful biological reactions. Last, after introducing targeting moiety, the polymer matrix should have an ability to bind specific sites to increase the residence time and improve the bioavailibity of drugs.
Herein, we synthesized a series of amphiphilic comb-like poly(oxyethylene)s with sulfide linkages to meet these criteria. Three polymers were prepared through polymer analogous reactions using poly(epichlorohydrin) as a starting polymer and Brij 76, 78, and mixtures of Brij 76 and poly(ethylene glycol) (Mn = 750) were substituted as side moieties, respectively. We introduced sulfide linkages in the side chain intentionally because they can form disulfide bond by self-crosslinking to enhance mucoadhesive adhesion1 and particle’s stability.
The polymer’s chemical structures and morphologies were studied using 1H NMR, Differential Scanning Calorimetry, Polarized Optical Microscopy, FT-IR spectrophotometry, X-ray Diffraction, etc.
