Branched polyethyleneimine(PEI) was well known as high gene transfection agent, However, utilization of PEI as gene carrier was limited because of high cytotoxicity. To solve this problem, PEI was introduced to chitosan(CS) back bone by using coupling reaction. Also, hyaluronic acid(HA) was introduced for targeted CD44-receptor in breast cancer cell. Its structural analysis of PEI-grafted chitosan(CSPEI) was confirmed by 1H-NMR. In addition, ternary complex of pDNA/CSPEI/HA was prepared by electrostatic charge interaction. Its gel retardation assay showed that pDNA was completely condensed by CSPEI and HA. Moreover, pDNA was protected from DNase and released by heparin. Also, transfection assay showed that ternary complexes was effectively transfected more than binary complexes by targeting effect of HA. These results suggest that ternary complexes can enhance transfection efficiency by targeted CD44-receptor in breast cancer cell.(NRF-2014R1A2A1A10053027). |
