Surface modification and characterization of poly (D,L-lactic-co-glycolic acid) scaffold with fructose |

Poly (D,L-lactic-co-glycolic acid) (PLGA) scaffold was pretreated using Argon plasma, and then UV polymerized with acrylic acid (AAc). Fructosylated surfaces were then obtained by coupling a fructose to the AAc graft chains with the aid of a water-soluble carbodiimide (WSC) and N-hydroxysulfosuccinimide (sulfo-NHS). The surfaces of the modified PLGA scaffolds were characterized by Fourier transform infrared spectroscopy-attenuated total reflectance (FTIR-ATR), electron spectroscopy for chemical analysis (ESCA) and atomic force microscopy (AFM). The hydrophilicity of the modified PLGA scaffolds was measured by a contact angle goniometry. The FTIR-ATR spectra of the surface-modified scaffolds showed the characteristic bands of PAAc and fructose. The ESCA survey scan, as well as carbon 1s (C1s), oxygen 1s (O1s), and nitrogen 1s (N1s) core spectra of the modified surfaces, confirmed the changes in chemical composition of the PLGA surfaces due to immobilized moieties. AFM images revealed that the surface roughness of the PLGA scaffolds increased after graft copolymerization with AAc, but did not change appreciably with the subsequent immobilization of the fructose ligands. Surface tension of the scaffolds increased after surface modification, and this result indicated that the hydrophilicity of the surface has improved significantly. |
