We report new bioreducible cystamine bisacrylamide(CBA)-amine polymers as novel gene delivery carriers. Disulfied bonds in the backbone of CBA-amine polymer can be cleaved to release the genetic materials from polyplexes into cytoplasm, where higher concentration of glutathione (GSH) exists. We have synthesized and screened a library of 38 different bioreducible CBA-amine polymers by gene transfection in vitro. We have identified the lead CBA-amine polymer (CBA-106) with an extremely high transfection efficiency by inducing GFP and Luciferase expression on mouse embryonic fibroblast (MEF) cells. The stable complexation between polymer and pDNA was confirmed by gel retardation assay, AFM imaging and their size was analyzed, in which, they were in the range of 200~500 nm by DLS measurement. We also have tested the viability of transfected cells and the results were significantly higher than that of conventional gene carrier such as PEI. These results clearly indicated that CBA-amines polymers have significant potential to be used as a promising gene delivery carrier. |
