제목 Fabrication and Evaluation of Hyaluronic acid-combined ternary complexes for targeted gene delivery against CD44-receptor overexpressed cancer cell
초록 Branched polyethyleneimine (bPEI) with many amine group was a high gene transfection agent. However, its utilization as gene carrier was limited because of high cytotoxicity. To overcome these obstacle, chitosan was introduced to bPEI by carbonyldiimidazole (CDI) as coupling agent. The chemical structure of chitosan-g-bPEI (CSPEI) was analyzed by 1H-NMR. The ternary complexes were prepared by electrostatic charge interaction. The particle size and morphological property were confirmed by DLS and TEM. Moreover, transfection efficiency and cellular uptake showed that ternary complexes displayed to gene expression and cellular uptake greater than binary complexes. Additionally, to demonstrate targeting effect of ternary complexes, competition assay was accomplished in HCT116 cell. These results suggest that ternary complex is a superb gene carrier with targeting effect against CD44-receptor overexpressed cancer cell line. (No. NRF-2016H1D5A1910499)
