Biodegradable polymer scaffolds, calcium phosphates (CaP), and bone morphogenetic protein (BMP)-2 are used as useful components for effective bone regeneration. We report on nano-hydroxyapatite (n-HAp) immobilized porous poly(lactide-co-glycolide) (PLGA) microspheres that can controllably release BMP-2 for a long time period (over 1 month). Based on our approach, the benefits of bioactive n-HAp, such as excellent cell attachment/proliferation and osteoconductivity, can be fully expressed, because n-HAp is exposed on PLGA scaffold pore surfaces at a nano-level. Furthermore, the released BMP-2 plays a significant role for stem cell differentiation to osteoblasts. We demonstrated that the porous PLGA microspheres with BMP-2-releasing n-HAp displayed the excellent osteoconductive function of n-HAp as well as osteoinductive role of BMP-2. They provided favorable environments for enhanced in vitro formation of bone tissues, which was confirmed by the specific biomarkers of bone formation. |
