Organocatalyzed Synthesis of Block Copolypeptides by Ring Opening Polymerization of α-Amino Acid N-Carboxyanhydrides |

We report the synthesis of polypeptides utilizing N-Heterocyclic Carbenes (NHCs) organocatalysts. NHCs were found to be highly effective for the living ring opening polymerization of α-amino Acid N-carboxyanhydrides (NCA) under mild conditions. The NHCs were able to produce polypeptides with controlled molecular weights (MWs), narrow molecular weight distributions and well-defined end group derived from the primary amine initiator. Block polypeptides (Poly(ε-carbobenzoxy-lysine)-block-(γ-benzyl-glutamate)) with controlled MWs and compositions were prepared by sequential NHCs-catalyzed polymerization of the corresponding (S)-ε-carbobenzoxy-lysine-N-carboxyanhydride (Z-Lys NCA) and (S)-γ-benzyl-glutamate-N-carboxyanhydride (Bn-Glu NCA) monomers. 1H NMR analysis reveals that adjusting the initial monomer with primary amine molar ratio can directly control the block copolymer chain length and composition. The experimental MWs were determined by gel permeation chromatography. |
