Aggregation of water-soluble paclitaxel probed by excimer fluorescence of pyrenebutyric acid tag |

We have investigated the aggregation property of a new water-soluble paclitaxel (PP7) tagged with fluorescent probe (pyrenebutyric acid, PBA) in water solution. From the steady-state fluorescence emission spectra, we have found that the excimer fluorescence of PBA tagged at PP7, centered at 470 nm, starts to appear at the concentration of 0.30(5) mM. This critical aggregation concentration of PBA-tagged PP7 is found to be much lower by about 3000-fold than that of free PBA. The appearance of the excimer fluorescence at such a low concentration indicates that the intermolecular interaction between the large hydrophobic parts of the paclitaxels effectively brings the fluoresent probes into proximity. The excimer fluorescence intensity and lifetime of PBA tagged at PP7 can be employed as useful probes for the further investigation into medical actions of this new water-soluble paclitaxel prodrug. |
