제목 | Preparation of Cell Adhesive Pluronic-META Derivatives Exhibiting Temperature-Sensitive Sol-Gel Transition Behaviors |
초록 | Pluronics are nontoxic poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) (PEO-PPO-PEO) triblock copolymers which form thermoreversible gels at high concentrations. These gels have received increasing attention over many years as drug delivery systems for topical, transdermal, ophthalmic and implantable applications and biomaterials.1,2 In particular, Pluronics F127 has been most widely used as proper block copolymers in pharmaceutical systems. For thermal behaviors in specific temperature, we have already synthesized functional Pluronic derivatives such as Pluronic-META (4-methacryloxyethyl trimellitic anhydride). In the present study, we have newly designed and synthesized temperature-sensitive and cell adhesive triblock copolymers that Arg-Gly-Asp (RGD) peptide ligand is grafted to Pluronic-META derivatives to obtain Pluronic-META-RGD (Fig. 1). The chemical structures of the Pluronic-META-RGD block copolymers were analyzed by FTIR, GPC, and 1H and 13C NMR. Sol-gel transition and particle size of the resulting Pluronic-META-RGD copolymers were characterized by tube tilting method (tilted the vial with 90˚ for 10 minutes) and light scattering, respectively. In addition, the interactions between these functional copolymers containing RGD ligand and cells are discussed for biomedical applications. Fig. 1. Synthesis scheme of Pluronic-META-RGD.
