Recently, quantum sized carbon-dot(C-dot), which consists of carbon and oxygen, passivated by polymer is reported and it shows excellent optical properties as well as environmentally, biologically good compatibility. Therefore, C-dot is suggested as an alternative of quantum dots(Q-dots) for biomedical applications to overcome Q-dots toxicity. Herein, we proposed C-dot-polyethylenimine(PEI) nanohybrid which shows not only high fluorescence but also complexation ability with plasmid DNA(pDNA). Specifically, the fluorescence of C-dot is quenched by gold nanoparticle(AuNP) which is known as super quencher, and AuNP surface is modified with positive charged PEI to make complex with pDNA. By dissociating pDNA with C-dot and AuNP, the quenched fluorescence of carbon-dot is recovered. Hence, gene delivery efficiency as well as real-time monitoring of association/dissociation of pDNA with its carrier will be studied, which is essential to understand the mechanism of non-viral gene delivery. |
