제목 A drug loaded QD-aptamer conjugate as a cancer imaging and therapeutic agent
초록 Here in we report a multimodal vehicle comprising of nanocrystal as imaging agent, aptamer as the targeting ligand and doxorubicin an anticancer drug. We hypothesize that after dox intercalation to A10 PSMA aptamer coupled to the CdSe/ZnS core-shell quantum dot (QD), the photoluminescence (PL) of QD gets quenched and the system is in a turn off state, and a dark quenching effect would be seen from dox as it remains bound to aptamer, and subsequently turned on by an increase in PL of QD and Dox as well due to the release of dox bound to the aptamer conjugated QD after uptake and delivery into the PSMA positive cells. In conclusion through in situ PL modulation of QD-aptamer by addition of Dox forming a physical complex with aptamer and showed targeting efficacy specifically to PSMA expressing LNCaP cells in vitro. Our data also demonstrated that QD-aptamer-Dox conjugate has equipotent efficacy as free Dox.
