제목 Heparin-based Self-assembled Nanoparticle for Photodynamic Therapy
초록 Photodynamic therapy (PDT) has been proposed as a useful theragnostic tool for cancer treatment. They generate highly reactive oxygen species when exposed to light and become highly cytotoxic. In spite of this interesting property of the photosensitizers as a therapeutic agent, their hydrophobicity has been the main cause of skin photosensitivity, which is the major side effect after PDT. Therefore, in this study, we prepared heparin/poly(β-benzyl-L-aspartate) (HP) amphiphilic conjugates and encapsulated pheophorbide-a (Pheo), one of the second generation PDT agents, into a self-assembled nanoparticle system of HP conjugates. Highly hydrophobic Pheo was successfully incorporated into HP nanoparticles during dialysis, and the drug-loading content depended on the feed weight ratio of drug. In the in vitro cell studies, the Pheo-loaded HP nanoparticles showed good cellular uptake and marked photocytotoxicity after light treatment, causing significant damage to cancer cell.
