Stem cell implantation is known to be a promising strategy for treating ischemic diseases. The major obstacle of stem cell transplantation, however, is stem cell viability. We proposed an method for preparing extracellular matrix (ECM) maintained stem cell to improve the engraftment of the transplanted stem cells. We synthesized pH-sensitive polymer and cultured human adipose derived stem cells (hADSCs) on the modified poly amino ester (mPAE) coated tissue culture plates (TCPs). The cells cultured on the mPAE coated TCPs and detached with low pH solution showed no statistical differences in vitro experiment compared to those of stem cells cultured on normal TCPs. Furthermore, the blood perfusion rate, microvessel density, and blood vessel regeneration in mPAE group were higher than those of no treatment, and TCP groups in vivo mouse model. The stem cell harvesting method with mPAE could be considered as an alternative for using conventional proteolytic enzyme treatment. |
