Enhancement of circulation time in vivo by surface modification of liposomes with ODA-grafted-comb polymer |

To enhance the stability of liposome in serum and prolong the circulation time in bloodstream, a number of liposomes with various functionalities have been developed. Thus, we prepared the octadecylacrylate-grafted-comb polymer (OGC) conjugated liposomes (OGC-liposomes). The comb polymer was synthesized by radical polymerization and composed of octadecylacrylate (ODA) backbone as a hydrophobic anchor and short poly(ethylene oxide) side chains as a hydrophilic chain. The stability of OGC-liposomes was evaluated by measuring the amount of adsorbed protein in serum using Bradford assay. Doxorubicin encapsulated OGC-liposomes was prepared by remote loading method and its circulation time in blood, biodistribution and antitumor activity were evaluated after intravenous injection in mice. The OGC-liposomes showed higher stability than other liposomal groups. The doxorubicin encapsulated OGC-liposomes showed more prolonged circulation time than those of conventional and PEG-coated liposomes. Additionally, antitumor activity of OGC-liposomes was enhanced, comparing with other liposomal formulation groups. |
