Model oligopeptide drug delivery into osteroclast |

The intracellular delivery of exogenous proteins into living cells with the help of membrane-permeable carriers has gained much attention in recent years. In this research, Nanoparticels functionalized with dendritic guanidine and amine groups at the surface were designed respectively for the delivery of osteoclast inhibiting peptides drug into cells. FACS and spectrofluorimetry analysis showed all nanoparticles were uptaken by cells. In addition, nanoparticles functionalized with dendrtic guanidine groups demonstrate better ability to cross the cell membranes. Time course of uptake results suggested that the fluorescence intensity of drugs increased with time. Nanoparticles functionalized with dendritic guanidine groups entered into cells more faster than those functionalized with dendritic amine groups or naked drug. All carriers exhibited no significant cytotoxicity against osteoclast. |
