In adoptive T cell therapy, the activity of cells cultured in vitro is often reduced and the transplanted cells may not stay in place long-term. Therefore, there is a need for development of a novel anti-tumor immunotherapy platform to deliver anti-cancer T cells in a stable and biologically active form. Given that, we prepared biocompatible nanofibers as a cell delivery carrier by using electrospinning method. To improve CD4 T cell isolation from spleen, we immobilized anti-CD4 antibody on the prepared nanofibers. we then measured CD4 T cell isolation efficiency and investigated the effect of immobilized anti-CD4 antibody on proliferation and release of CD4 specific T cells. The results showed that the antibody-immobilized scaffold was effective in capturing CD4 T cells. Thus, our nanofiber platform may be able to isolate CD4 T cells and to maintain anti-cancer immune cell activity which can be further used in adoptive T cell therapy. |
