To maximize the therapeutic efficacy in gene delivery, stimuli-responsive delivery carrier has been developed in the sense that it controls the cytosolic conveyance and rate of gene release. We present herein a hybrid nanocomposite composed of single layered MoS2, polyethylenimine (PEI), and polyethylenglycol (PEG). The PEI and PEG were conjugated in a reducible manner, disulfide bond, releasing them in reducible cytosolic environment. Photothermal conversion of MoS2 nanosheet was employed in order to induce photothermally trigged endosomal escape upon the near infrared laser subjection. After endosomal escape, intracellular reducing environment let the polymers detach, facilitating the gene release from the nanocomposite. The sequential process initiated by external and internal stimuli remarkably ehnaced gene delivery efficiency by effective endosomal escape and gene release without severe cytotoxicity. |
