Efficient Delivery of Therapeutic Oligonucleotides based on Nanotechnology |

Even though many nanomaterials enjoy successful demonstrations in bioapplications, lack of biocompatibility and high cytotoxicity often become hurdles for practical bioapplications. On the other hand, it is important to achieve enough efficiency based on chemically well-defined systems with efforts to understand mechanism at molecular level. Here, we develop biocompatible nanomaterials to efficiently deliver oligonucleotides including small interfering RNA (siRNA) and DNAzyme to mammalian cells to induce sequence-dependent target gene silencing. We demonstrated our strategy in two disease models— cancer and hepatitis C— with iron oxide nanoparticles and porous silica nanoparticles. The nanocomplex loaded with oligonucleotides showed high knockdown efficacy with lower cytotoxicity than Lipofectamine 2000 (conventional transfection reagent) while not evoking undesired immune responses in vitro. |
