Cleavable PEG Nanogels encapsulating DNA |
초록 |
Water soluble cationic polymers have been widely used to form polyelectrolytic complex which can deliver genes to the cell. Here we report for the first time that plasmid DNA can be physically encapsulated within cross-linked Poly(ethylene glycol)(PEG) nanogels. DNA was encapsulated within PEG nanogel by interacting multi-armed –SH functionalized PEG to DNA through hydrogen bond in organic solvent. PEG nanogel was linked with disulfide bond and DNA could be released in high GSH concentration. Average size of nanogel is 200~400nm and the size was dependent on molar ratio of PEG-SH and crosslinker. PEG nanogel could be used as an excellent gene carrier in vitro and in vivo. |
