Dermal-epidermal junction-targeted transdermal delivery using phase modulus-tuned, cell penetrating peptide-tagged polymeric nanocarriers |

We establish a new transdermal delivery system in which drug molecules encapsulated in polymeric micelles are delivered to dermal-epidermal junction (DEJ). For this, phase modulus-regulated polymeric micelles were fabricated via co-assembly of hybrid amphiphilic triblock copolymers, poly (ethylene oxide)-b-poly (ε-caprolactone)-b-poly (ethylene oxide) and poly (ethylene oxide)-b-poly (propylene oxide)-b-poly (ethylene oxide). The modulus of micelle phase could be controlled depending on the composition of triblock copolymers. To improve transdermal delivery, in vitro cell-penetration study was carried out using the polymeric nanocarriers conjugated with cell-penetrating peptide. Furthermore, we showed that the hybrid polymeric nanocarriers with well-tuned phase modulus as well as specific binding ability diffused readily into the stratum corneum and reached richly to the area of DEJ. |
