제목 Enhanced gene silencing effect of oligopeptide-modified chitosan/siRNA nanoaprticles
초록 RNA interference (RNAi) is a conserved mechanism of gene silencing and has shown potential for the treatment of various diseases, including cancer and viral infection. However, the delivery of small interfering RNA (siRNA) should overcome significant obstacles for in vivo applications due its limited stability. Thus appropriate delivery carriers are required for successful siRNA delivery. We hypothesized that chitosan modified with positively charged nonaaginine residues could enhance the stability chitosan/siRNA complexes and the insertion of spacer arm between chitosan and nonaarginine residues could improve cellular association and transfection efficiency. Various physicochemical properties of oligopeptide-modified chitosan/siRNA nanoparticles were investigated and their in vivo therapeutic efficacy was tested using a mouse model. This approach to developing a novel delivery system using natural polymers may provide a useful tool to many gene delivery applications.
