In this study, we developed E. coli strain that is able to utilize formate as a carbon source in sugar-free conditions, through the introduction of the serine cycle pathway into E. coli. For formate assimilation, the genes related to the tetrahydrofolate cycle and serine cycle were overexpressed from Methylobacterium extorquens AM1. Additionally, adaptive laboratory evolution (ALE) was carried out until the desired phenotypes emerged to overcome the problems of toxicity and low biomass conversion rate of formate. After conducting ALE experiment for five months, mutant strains that can g format as a carbo source desired phenotype was screened out and its genome was sequenced. Based on genome sequencing, a few possible mechanisms that might increase the formate utilization were examimed. Finally, the resulting E. coli was engineered to convert formate into ethanol. |
