In the present study, alginate nanoparticles containing biotin conjugated exendin-4 were formulated and evaluated different dissolution rate in various pH, an GI enzyme-resistant, superior intestine absorptive for improved peroral delivery. Peptide surface modification with biotin (vitamin H), which actively traverses the intestine membrane via sodium-dependent multivitamin transport (SMVT), was chosen as a promising approach. Alginate was also selected as the nanoparticle-forming material based on its anionic property and mucoadhesiveness, which protect exendin-4 from degradation by gastric juice and allow it to be slowly released in the ilem or the upper intestine. |
