This works introduce shell cross-linked protein/QD hybrid nanocapsules as an efficient nanocarrier for tumor-targeted bio-imaging. Cadmium-metal-free CIS/ZnS core-shell QDs are synthesized using a hot-injection method. The QDs are mixed with NHS-activated six-armed PEG in an organic solution, and then emulsified into an aqueous solution containing human serum albumin (HSA). The 6-armed PEG readily diffuses out toward the aqueous phase. At the oil/water interface, the PEG chains react with the adsorbed HSA and cross-links the proteins to form a protein-PEG network, resulting in shell cross-linked nanocapsules incorporating the QDs. The prepared hybrid nanocapsules show excellent dispersion stability in a serum-containing medium. The feasibility of tumor-targeted in vivo bio-imaging is demonstrated using a mouse model injected with tumor cells after tail vein injection of the nanocapsules.
