Bio-Inspired Self-Assembly of Peptide Dendrimers into Capsid-Like Nanostructure for Drug and Gene Delivery |

Supramolecular self-assembly of dendrimers is a promising approach to engineer advanced vectors for therapeutic agent delivery. Peptide dendrimers coupled with linear peptides spontaneously assembled into well-defined capsid-like nanstructure in water, whose size and morphology were controllable. The capsid-like nanstructure could serve as a pH-responsive nanovector for anticancer drug delivery due to inherent features of peptide-based materials.The smart nanovector significantly enhanced antitumor efficacy of hydrophobic doxorubicin against HepG2 human hepatocyte tumor cell line and 4T1 murine breast tumor model. More importantly, a family of capsid-like nanovectors were used to develop towards artificial virus for gene delivery. These artificial viruses achieved high gene transfection efficiency both in vitro and in vivo gene delivery.As our continuous interest in self-assembly of peptide dendrimers, we are developing more strategies for constructing novel supramoleculardendrimers to satisfy various biomedical applications, such as multi-drug delivery, drug/gene co-delivery and theragnosticagent delivery. |
