The amine-lipid based lipomers for mRNA delivery into MEF cells |

Delivery of messenger RNAs (mRNAs) has drawn much attention since it show great advantages over conventional plasmid DNA (pDNA) delivery. We have developed new amine-lipid based lipomers for efficient delivery of mRNA to mouse embryonic fibroblast (MEF) cells. Resulting lipomers show facile self-assembly with negatively charged mRNA through electrostatic interaction. A library of eight different amine-lipid based lipomers has been screened by in vitro gene transfection. As a result, amine-epoxydodecane (102-C10) show excellent delivery of mRNA and thereby express high Green Fluorescence Protein (GFP) expression in MEF cells. In addition, the viability of tansfected cells is well maintained using a 102-C10 lipomer. These results indicate that amine-lipid based lipomers have significant potential as a promising mRNA delivery carriers. |
