Robust and highly sensitive detection of pathogens in latent period is essential to prevent widespread of infectious diseases. In the recent years, Middle East Respiratory Syndrome (MERS) has been spread out over the world. It was highly infectious and made numerous patients. It is getting more important to have early diagnosis to stop pandemic spread out of disease. In this study, facile and robust microfluidic array chip assay named as ‘DhITACT (DNA hydrogel formation by isothermal amplification of complementary target)’ has been developed based on rolling circle amplification (RCA) of adaptor templates upon the hybridization of MERS pathogens. Long single strands of RCA products can be visualized by SYBR gold staining. When a biological sample containing MERS pathogen is applied to the DhiTACT, RCA of the DNA template can be achieved and can be visualized by under a UV lamp. In contrast, if there is no MERS pathogen in sample, no fluorescence signal can be seen. It is likely that the development of facile and sensitive MERS pathogen detection assay may relieve excessive workloads. |
