제목 Collagen-bone sialoprotein coating on BCP scaffold for enhanced mineralization
초록   Bone sialoprotein (BSP) is a non-collagenous acidic glycoprotein that is highly expressed in mineralized tissues. Biological mineralization is important because it contributes to the strength and rigidity of bone. BSP may have a specific role in enhancing apatite nucleation and mineralization. So, triple-helical collagenⅠ(rat tail COLⅠ)-BSP was coated on fabricated biphasic calcium phosphate (BCP) scaffolds. First, the scaffolds were prepared from BCP as basic materials by extrusion process. BCP filaments filled with carbon were cut to cylindrical particles. These particles were compressed in die and sintered to remove carbon. Cylindrical BCP scaffolds with macro pits were fabricated. Then, COLⅠ and BSP were coated on these scaffolds. Characterization and material properties of these scaffolds were evaluated based on scanning electron microscopy (SEM) observation, X-ray diffraction (XRD) and Fourier transform infrared (FTIR) spectra. BSP release profile was also determined. Cell viability on these scaffolds was examined by in vitro experiment using pre-osteoblast MC3T3-E1 cells. Compared with non-coated BCP scaffolds, the BSP-COLⅠ coated BCP scaffolds exhibited excellent biocompatibility, cell proliferation and cell adhesion behavior as determined by MTT assay and immunofluorescence analysis using confocal microscopy. Based on these results, BSP-COLⅠ coated BCP scaffolds could be considered as potential system for dental, bone and tissue engineering applications in enhancing biological mineralization.
