제목 In-vitro Biological and in-vivo Evaluation of Gelatin-Pectin-BCP Porous Scaffold for Bone Tissue Engineering
초록 A biodegradable composite porous scaffold comprising Gelatin (GE), Pectin (P) and Biphasic Calcium Phosphate (BCP) nanoparticles matrix was developed and studied for bone tissue engineering. The aim of this study was to prepare three-dimensional scaffolds using these two natural polymers and to add BCP for bone regeneration. The role of BCP nanoparticles in scaffolds is to enhance the osteoblast induction in-vitro and newly-bone formation in-vivo. Scanning electron microscopy, porosity, Differential scanning calorimetry and X-ray diffraction were used to examine the physical properties of the scaffolds. To assay the cytocompatibility and cell behavior on the GE-P-BCP porous scaffold, MTT assay, immunefluorescence and osteogenesis analysis using pre-osteoblast MC3T3-E1 cells were conducted. In-vivo study was evaluated by micro-CT, hematoxylin&eosin and immunohidtochemistry (OPN, OCN, COL I and COL II) on rabbit models. Our results provide a novel and simple method to obtain an adequate scaffold for osteoblast cells and indicate that GE-P-BCP porous scaffold could be a good candidate for bone tissue engineering.
