In vitro recognition of breast cancer cells using herceptin-conjugated microdroplets |

In this study, we report an recognition in which antibodies are conjugated on the surface of liquid crystal(LC) droplets rather than on solid surfaces. 4-Cyano-4´-pentylbiphenyl(5CB, nematic liquid crystal) microdroplets were prepared by using O/W emulsion system. Amphiphilic diblock copolymer of poly (styrene)-b-poly (acrylic acid) (PS-b-PA) was used as a surfactant to prepare the emulsion. The hydrophobic styrene block is miscible with the hydrophobic 5CB and incorporates in the micelle core. The hydrophilic PA block is in the outer shell. Herceptin was subsequently conjugated to the PA block to preferentially target SK-BR-3 cells (human breast cancer cells) that have HER2 receptors on the cell membrane. The configuration of the LC (5CB) in the emulsion was changed as a result of antigen-antibody interaction forces. This observation thus demonstrates potentiality of the antibody-conjugated LC emulsions as a biosensor for specific cell and protein detection without additional steps. |
