In this study, a fed-batch culture process of recombinant Escherichia coli for the production of recombinant urate oxidase incorporated with unnatural amino acid, 4-azido-phenylalanine(AzF). Firstly, through a series of flask cultures of 10 types of recombinant E. coli, which were transformed with two plasmids of pEVOL_pAzF and pQE80_UoxR for the expression of urate oxidase incorporated with Azf (UO_Azf), recombinant E. coli Top10 was selected as the best strain for the production of UO_Azf. Using a recombinant E. coli Top10 strain, a series of fed-batch cultures were conducted by varying several variables such as the IPTG induction time, the amount of glucose and complex nitrogen sources fed per feeding and the Azf concentration. As a result, it was confirmed that the specific activity of UO_Azf increased to a maximum of 41.14 U/mg when the culture was operated at the best condition. |
