Fabrication and Characterization of the Multiple Times Silk Fibroin Coated Micro-Channeled BCP Granules and its in-vitro Evaluation for Improved Bone Regeneration |

This study describes the fabrication and characterization of silk fibroin coated micro-channeled BCP granules. 10% (w/v) silk fibroin solution has been coated on BCP granules. Silk-coated BCP granules were prepared by immersing the ceramic granules in silk solution and pipetting the solution through the granules to ensure the uniform infiltration. The coating was stabilized by immersing the silk coated granules in methanol for 5 minutes to induce β-sheet formation, followed by vaccum drying at 800 C for 10 minutes. This coating procedure was repeated 1, 5 & 7 times to give silk-coated BCP granules with different coating thickness. The surface morphology of silk coated granules has been observed by SEM. The binding energy of both only granules and silk fibroin coated granules were measured by XPS. FTIR and XRD were performed in order to confirm the silk fibroin presence in the granules. MCT3-E1 pre-osteoblasts cells were seeded on the only granules and silk coated granules for conduction in-vitro study. MTT assay, live-dead assay and cell proliferation behaviour were observed as an in-vitro study in order to confirm the biocompatibility of silk coated BCP granules for applying in bone tissue engineering. |
