The rational design of integrated, multi-functional polymeric carriers is prerequisite for the successful nonviral cancer gene therapy. In this study, we developed identical functionalization conditions which would allow simultaneous installation of various types of functionalities in one-pot reaction under mild conditions through pre-thiolation strategy incorporating thiol-containing components. Newly designed gene delivery system possessing multiple functional domains such as linear polyethylene glycol (PEG), cyclic NRG (CNGRCK, cNGR) tumor-targeted peptide and bioreducible branched polyethylenimine (BPEI) was synthesized and characterized. We also evaluated polymeric vectors with respect to the combining effect of targeting and intracellular reducible functions concerning gene transfection in various cancer cells with the view point of physicochemical properties and cellular uptake mechanism. |
