We report a smart nanovector, hyaluronic acid (HA)-coated on the surface of cationic polymers (imidazole-graft-poly-L-lysine, pLys-g-Im) polyplexes containing Bcl-xL specific shRNA (HA/pLys-g-Im/shRNA), for gastric cancer therapy. The prepared ternary polyplexes have the negative surface charge of -24 mV and the size of around 160 nm at N/P ratio of 5 and HA/pLys-g-Im molar ratio of 0.03. Furthermore, gel electrophoresis and cell viability tests demonstrated the ternary polyplexes showed high stability and no cytotoxicity due to the anchored HA molecules on the surface of pLys-g-Im/pDNA binary polyplexes. Consequently, selective cancer cell death was achieved by CD44-mediated gene delivery and the internalized gene was effectively escaped from endosomes due to buffering capacity of imidazole groups under acidic environment, indicating that the technique describe herein might allow selective destruction of metastatic gastric cancer. |
