Pancreatic islet transplantation is the ideal therapy for the treatment of type 1 diabetes mellitus (T1DM). However, transplanted islet grafts suffer oxidative stress and nonspecific inflammatory reactions. Bilirubin has showed anti-oxidative and anti-inflammatory functions. Although bilirubin has multi-potency as a cytoprotective agent, its clinical application has been limited due to its solubility. To overcome this limitation, hydrophilic PEG polymer was introduced to prepare PEGylated bilirubin nanoparticle (APBN) formed in aqueous media. In this study, we synthesized and evaluated the function of APBN. Administration of APBN (40 μM) effectively protected islet cells from oxidative stress and nonspecific inflammatory response in vitro. Furthermore, treatment of APBN could increas islet graft survival about 3-fold in vivo islet transplantation. Collectively, these results demonstrated the APBN would be a novel remedy for the successful outcome of islet xenotransplantation. |
