A new chemical tool for the identification of O-GlcNAc porteome, O-GlcNAz-modified proteins |
초록 |
We engineered a phosphine reagent by installing an easily cleavable disulfide linker between the phosphine skeleton and a biologically active tag such as His-tag or biotin. Using this reagent, O-GlcNAc-modified proteins (or peptides) are distinguished from proteins that are not O-GlcNAc-modified.In this strategy, tagged O-GlcNAc-modified proteins bind to functionalized magnetic nanoparticles allowing the O-GlcNAc bound nanoparticles to be easily isolated from the whole cell extract.Then, O-GlcNAz-modified proteins are easily released from the nanoparticle by breaking disulfide linkage of the engineered phosphine reagent.This new chemical tool will accelerate the identification of O-GlcNAc modified proteins and the site of modification. |
