Carotenoid and photosensizer dual-loaded polymeric micelles with controllable singlet oxygen production: preparation, characterization, singlet oxygen scavenging properties, and in vitro PDT efficacy |

In photodymamic therapy (PDT), the key cytotoxic agent is singlet oxygen (1O2) that is produced through the photochemical reactions between photosensitizer (PS) and the surrounding molecular oxygen under light irradiation. By precise control of the ability of PS to generate 1O2, the photodamage to non-targeted tissues can be effectively prevented. Here, we prepared β-carotene (CAR) and pheophorbide a (PhA) dual-loaded PEG-PCL micelles with a controllable phototoxicity. CAR is well known for its role in scavenging 1O2. This study aimed to evaluate whether incorporation of CAR and PhA into PEG-PCL micelles can silence the 1O2 generation prior to drug release at desired sites. It was confirmed that CAR can seriously diminish the 1O2 generation of PhA through directly scavenging 1O2. But, PhA remained its capacity for producing 1O2 when CAR and PhA were released from micelles. Moreover, dual-loaded micelles displayed efficient cellular uptake and effective PDT activation in tumor cells. |
