Polymeric Micelles for Multi-Drug Delivery, Optical Imaging and Theragnostics |

Polymeric micelles are a mature class of drug carriers, satisfying strict requirements for safety, drug solubilization and controlled release. PEG-b-PLA micelles solubilized paclitaxel, 17-AAG (Hsp90 inhibitor) and rapamycin (mTOR inhibitor) in water. IV injection of multi-drug PEG-b-PLA micelles “supercharged” paclitaxel, resulting in potent antitumor efficacy in A549 non-small cell lung and MDA-MB-231 breast xenograft models, co-targeting survival signaling pathways. In a LS180 colon carcinoma xenograft model, multi-drug PEG-b-PLA micelles enhanced the tumor accumulation of PEG-b-PCL micelles carrying a lipophilic near-infrared carbocyanine dye (DiR) by 2.1-fold, based on ex vivo optical imaging: Fluorescence tumor-to-muscle ratio of 149 with “tumor priming,” versus 68 for PEG-b-PCL micelles without pre-treatment. Current research focuses on a novel theragnostic strategy for ovarian cancer treatment, involving tumor priming, apoptosis targeting/optical imaging and image-guided surgery in an orthotopic ES-2 xenograft model. |
