Reactive oxygen species (ROS)-sensitive degradable polymer, poly-(1,4-phenyleneacetone dimethylene thioketal) (PPADT) has been used to protect siRNA from the low pH of the gastrointestinal tract and target its delivery to ROS-rich inflamed intestinal tissue (Nature Materials, (2010) 9, 923-928). This poster presents the synthesis of PPADT and subsequent PEGylation to form PPADT-PEG block copolymers. By varying the chain lengths of PPADT and PEG, we prepared PPADT-PEG block copolymers having various ratios of hydrophobic and hydrophilic blocks, which may affect the degradation behaviors of the block copolymer by ROS. The ratio of p-toluenesulphonic acid (PTSA; catalyst) to monomers were particularly important for fine tuning of molecular weight and polydispersity. |
