Effects of Transforming Growth factor-β/Low Molecular Weight Chitosan Conjugates on the Repair of Osteoarthritis in Vivo |

It has been reported that TGF-β is related to biological activity, such as chondrogenesis and cartilage specific matrix formation. However, its short half-life is a most mooted point in application as therapeutic agent for osteoarthritis, so we have attempted to increase its half-life by conjugating with LMC. In this study, we prepared TGF-β-LMC conjugates by using carbodiimide, and investigated its effects on articular cartilage defects of the rabbit's knees. Full thickness cartilage defect was made at both rabbit's knees, and after 3 days TGF-β-LMC conjugates was injected into one side and PBS was injected into the other side. Histological results of experimental group at 6 and 12 weeks postoperatively were superior to control group. The results of 12 weeks was more remarkable than 6 weeks postoperatively. By these results, it is suggested that TGF-β/LMC conjugates can be used as a therapeutic agent for Osteoarthritis. |
