The preparation of TGF-β-LMC conjugates and the effect on osteoarthritis in Vivo |

Transforming Growth Factor - β is most effective human growth factor to osteoarthritic knee. But it's short half-life and expensive costs were a most mooted point as therapeutic agent for osteoarthritis, so we attempted to increase its half-life by conjugation with low molecular chitosan by using EDC as a cross-linking agent. After preparation of TGF-β-LMC conjugates, the biologic effect of TGF-β-LMC on osteoarthritic articular cartilage was compared with pure TGF-β. 1ml of TGF-β-LMC conjugate(15ng/ml) or pure TGF-β(15ng/ml) were injected to the intra-articular space of the experimental osteoarthritic knee of the rabbit. The specimens were analysed by gross findings, histologic examinations, and histo-morphometric studies. In histologic examination, TGF-β-LMC conjugate group shows less severe osteoarthritic changes than pure TGF-β and control group. |
