Intracellular delivery of proteins to apply therapeutic purposes has received much attention in pharmaceutical field. For effective protein delviery, especially using nano-sized carrier, several limitations such as the denaturation of protein and low intracellular uptkae efficiency must be resolved. Heparin derivatives have an advantage as protein delivery carriers due to its binding affinity with protein. In this study, heparin-Pluronic (HP) nanogel was developed as an effective carrier for intracellular delivery. The chemical structure of HP copolymer was characterized by 1H NMR and FT IR. The mean size of the RNase A loaded HP (HPR) nanogel prepared by direct dissolution method was performed by DLS. The cell viability of HPR nanogel was investigated by MTT assay. HPR nanogel was internalized to the cytosol as well as the nucleus confirmed by confocal microscopy.
