Hyaluronic acid (HA) is a biodegradable, biocompatible, and non-immunogenic linear polysaccharide. A number of strategies for the chemical modification of HA through carboxyl and hydroxyl groups have been reported for various medical applications. The objective of this study was to develop degradation controlled novel HA derivatives for drug delivery and tissue engineering applications. HA has different conformational structure in water and in organic solvent, and the carboxyl group of HA is known to be the recognition site of hyaluronidase. Accordingly, HA was chemically modified in the mixed solvent of water and ethanol by grafting adipic acid dihydrazide (ADH) to the carboxyl group of HA, which contributed for sustained degradation of HA by hyaluronidase. Degradation of the HA derivatives was monitored by measuring the optical density of degradation product with double bonds after hyaluronidase SD treatment. Degradation controlled HA will be assessed in detail. |
