Restenosis and Thrombosis are still drawbacks of stent use; that is why many types of stents have been produced until today. Researchers have found that nitric oxide (NO) has anti-restenotic effects due to its role in keeping a healthy vasculature. In this study, we focused on trying to reach an optimum coating condition for liposome containing DETA-NONOate, a NO donor, on coronary stents, which will accomplish a long-term sustained release of NO by comparing three main coating techniques, direct coating on metal surface, direct coating on polymer layer, and layer-by-layer coating technique. Layer-by-layer coating accomplished a sustained release of NO for about 5 days with two drug release bursts when compared to the other two coating techniques. In addition, biological studies revealed that samples show significantly reduced fibrinogen adhesion and enhanced cell growth when compared to control due to the sustained release of NO and surface properties. |
